Temperate pastures are often managed with P fertilizers and N2-fixing legumes to maintain and increase pasture productivity which may lead to greater nitrous oxide (N2O) emissions and reduced methane (CH4) uptake. However, the diel and inter-daily variation in N2O and CH4 flux in pastures is poorly understood, especially in relation to key environmental drivers. We investigated the effect of pasture productivity, rainfall, and changing soil moisture and temperature upon short-term soil N2O and CH4 flux dynamics during spring in sheep grazed pasture systems in southeastern Australia. N2O and CH4 flux was measured continuously in a High P (23 kg P ha?1 yr?1) and No P pasture treatment and in a sheep camp area in a Low P (4 kg P ha?1 yr?1) pasture for a four week period in spring 2005 using an automated trace gas system. Although pasture productivity was three-fold greater in the High P than No P treatment, mean CH4 uptake was similar (?6.3?±?SE 0.3 to ?8.6?±?0.4 μg C m?2 hr?1) as were mean N2O emissions (6.5 to 7.9?±?0.8 μg N m?2 hr?1), although N2O flux in the No P pasture did not respond to changing soil water conditions. N2O emissions were greatest in the Low P sheep camp (12.4 μg?±?1.1 N m?2 hr?1) where there were also net CH4 emissions of 5.2?±?0.5 μg C m?2 hr?1. There were significant, but weak, relationships between soil water and N2O emissions, but not between soil water and CH4 flux. The diel temperature cycle strongly influenced CH4 and N2O emissions, but this was often masked by the confounding covariate effects of changing soil water content. There were no consistently significant differences in soil mineral N or gross N transformation rates, however, measurements of substrate induced respiration (SIR) indicated that soil microbial processes in the highly productive pasture are more N limited than P limited after >20 years of P fertilizer addition. Increased productivity, through P fertilizer and legume management, did not significantly increase N2O emissions, or reduce CH4 uptake, during this 4 week measurement period, but the lack of an N2O response to rainfall in the No P pasture suggests this may be evident over a longer measurement period. This study also suggests that small compacted and nutrient enriched areas of grazed pastures may contribute greatly to the overall N2O and CH4 trace gas balance. 相似文献

Cre recombinase (Cre)-mediated targeted insertion of a transgene is a powerful technique that can be used to tailor genomes. When combined with somatic cell nuclear transfer it could offer an efficient way to generate transgenic livestock with site-specific genetic modifications that are free of antibiotic selection markers. We have engineered primary bovine fibroblasts to contain a chromosomal acceptor site with incompatible loxP/lox2272 sites for Cre-mediated cassette exchange and show for the first time that Cre-mediated targeting can be applied in these acceptor cells. Molecular characterization of the resulting cell clones revealed Cre-mediated transgene insertion efficiencies of up to 98% when antibiotic selection was used to identify transgene containing cell clones. Most clonal lines also contained random insertions of the targeting and Cre expression plasmids with only about 10% of the clones being exclusively modified by the intended targeted insertion. This targeting efficiency was sufficient to enable the isolation of correctly targeted clones without the help of antibiotic selection. Therefore, this recombinase-mediated insertion strategy has the potential to produce transgenic cattle from antibiotic selection marker-free somatic cells with transgenes inserted into proven genomic loci ensuring reliable expression levels. 相似文献

Raspberry volatiles are important for perceptions of sensory quality, mould resistance and some have nutraceutical activities. Twelve raspberry character volatiles were quantified, 11 of them in fruit from two seasons, from plants from the Glen Moy × Latham mapping population growing in both open field and under cover (polytunnels). Effects of season and environment were examined for their impact on the content of α-ionone, α-ionol, β-ionone, β-damascenone, linalool, geraniol, benzyl alcohol, (Z)-3-hexenol, acetoin, acetic and hexanoic acids, whilst raspberry ketone was measured in one season. A significant variation was observed in fruit volatiles in all progeny between seasons and method of cultivation. In this article I argue that the proper subjects of intangible property claims include medical records, genetic profiles, and gene enhancement techniques. Coupled with a right to privacy these intangible property rights allow individuals a zone of control that will, in most cases, justifiably exclude governmental or societal invasions into private domains. I argue that the threshold for overriding privacy rights and intangible property rights is higher, in relation to genetic enhancement techniques and sensitive personal information, than is commonly suggested. Once the bar is raised, so-to-speak, the burden of overriding it is formidable. Thus many policy decisions that have been recently proposed or enacted – citywide audio and video surveillance, law enforcement DNA sweeps, genetic profiling, national bans on genetic testing and enhancement of humans, to name a few – will have to be backed by very strong arguments. 相似文献

The combined effects of endurance run training and renal hypertension on cytosolic Ca2+ concentration ([Ca2+]c) dynamics and Na+-dependent Ca2+ regulation in rat left ventricular cardiomyocytes were examined. Male Fischer 344 rats underwent stenosis of the left renal artery [hypertensive (Ht), n = 18] or a sham operation [normotensive (Nt), n = 20]. One-half of the rats from each group were treadmill trained for >16 wk. Cardiomyocyte fura 2 fluorescence ratio transients were recorded for 7 min during electrical pacing at 0.5 Hz, 2 mM extracellular Ca2+ concentration, and 29 degrees C. The rate of [Ca2+]c decline was not changed by run training in the Nt group but was reduced in the Ht group. At 7 min, cardiomyocytes were exposed to 10 mM caffeine in the absence of Na+ and Ca2+, which triggered sarcoplasmic reticular Ca2+ release and suppressed Ca2+ efflux via Na+/Ca2+ exchanger. External Na+ was then added, and Na+-dependent Ca2+ efflux rate was recorded. Treadmill training significantly enhanced Na+-dependent Ca2+ efflux rate under these conditions in the Nt group but not in the Ht group. These data provide evidence that renal hypertension prevents the normal run training-induced modifications in diastolic [Ca2+]c regulation mechanisms, including Na+/Ca2+ exchanger. 相似文献
